Abstract:
Miniaturization related to the reduction to the micrometer scale of lab on chip devices considerably increases the throughput of bioassays or chemical reactions. One aim is to fabricate microfluidic chips in integrated platforms for Micro-Total Analysis System applications. Microfluidic platforms are attractive tools for cell biology as they offer accurate control of cellular environment and can be also used for analyzing sub-cellular content at the single-cell level.
Herein we propose laser fabrication of functional biochips for human cell testing. Hybrid complex glass and polymeric 3D configurations were fabricated by femtosecond lasers for a specific biological application. Concretely, we designed and fabricated biomimetic scaffolds for study of cancer cells inside glass micro-channels. Femtosecond laser assisted wet etching of a photosensitive glass followed by two-photon polymerization of a negative epoxy-resin allowed the size lowering to smaller details, to cell dimension or below. Time lapse imagining of cancer cells was carried out inside biochip during different time intervals. The cells were found responsive to chemo-gradient concentration created through 2 µm diameter channels of the 3D scaffold.
